How and why is the bacterium Thermus aquaticus employed in recombinant DNA technology? Explain. |

Answer» DNA polymerase (Taq polymerase) in recombinant technology is obtained from a bacterium, Thermus aquaticus. (i) DNA polymerase (thermostable) remain active during the high temperature induced during denaturation of double stranded DNA. (ii) This enzyme extends the primers using the nucleotides provided in the reaction and the genomic DNA as template. (iii) The repeated amplification is achieved by this enzyme and the amplified fragment if desired can be used to ligate with a vector for further cloning. |
